Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002528.7(NTHL1):c.98G>A (p.Arg33Lys), citing Ambry Variant Classification Scheme 2023: The p.R41K variant (also known as c.122G>A), located in coding exon 1 of the NTHL1 gene, results from a G to A substitution at nucleotide position 122. The arginine at codon 41 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
