Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.1051A>G (p.Lys351Glu), citing Ambry Variant Classification Scheme 2023: The p.K351E variant (also known as c.1051A>G), located in coding exon 10 of the TSC2 gene, results from an A to G substitution at nucleotide position 1051. The lysine at codon 351 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
