Uncertain significance for Fanconi anemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000135.4(FANCA):c.4088A>G (p.Lys1363Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine with arginine at codon 1363 of the FANCA protein (p.Lys1363Arg). The lysine residue is moderately conserved and there is a small physicochemical difference between lysine and arginine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has not been reported in the literature in individuals with FANCA-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt FANCA protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
