This variant lies in the AIPL1 gene (transcript NM_014336.5) at coding-DNA position 119, where T is replaced by A; at the protein level this means replaces methionine at residue 40 with lysine — a missense variant. Submitter rationale: This sequence change replaces methionine, which is neutral and non-polar, with lysine, which is basic and polar, at codon 40 of the AIPL1 protein (p.Met40Lys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of retinitis pigmentosa (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1019070). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
