NM_004260.4(RECQL4):c.2050A>C (p.Thr684Pro) was classified as Uncertain significance for Baller-Gerold syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with proline at codon 684 of the RECQL4 protein (p.Thr684Pro). The threonine residue is weakly conserved and there is a small physicochemical difference between threonine and proline. This variant is not present in population databases (ExAC no frequency). This variant has been observed in an individual with breast cancer (Invitae). However, in that individual a pathogenic allele was also identified in RECQL4, which suggests that this c.2050A>C variant was not the primary cause of disease. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. The proline amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
