Uncertain significance for Cystic fibrosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000492.4(CFTR):c.4028G>A (p.Gly1343Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with aspartic acid at codon 1343 of the CFTR protein (p.Gly1343Asp). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and aspartic acid. This variant is present in population databases (rs773458471, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with CFTR-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
