NM_020297.4(ABCC9):c.3131G>A (p.Gly1044Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G1044E variant (also known as c.3131G>A), located in coding exon 25 of the ABCC9 gene, results from a G to A substitution at nucleotide position 3131. The glycine at codon 1044 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
