NM_033629.6(TREX1):c.95C>T (p.Thr32Met) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: TREX1 c.95C>T (p.Thr32Met) results in a non-conservative amino acid change located in the Exonuclease, RNase T/DNA polymerase III domain (IPR013520) of the encoded protein sequence. Four of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 251328 control chromosomes (gnomAD). c.95C>T has been reported in the literature in at-least one individual affected with early onset high myopia (eoHM) who underwent exome sequencing (eoHM) (Zhou_2018). It was also found in the heterozygous state in individuals affected with Aicardi Goutieres Syndrome 1 (Wang_2020, Gorkumez_2023). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 29453956, 33042144, 36964972). ClinVar contains an entry for this variant (Variation ID: 1018555). Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
