Uncertain significance for Nephrolithiasis/nephrocalcinosis — the classification assigned by Ambry Genetics to NM_000388.4(CASR):c.1894G>A (p.Gly632Ser), citing Ambry Variant Classification Scheme 2023: The p.G632S variant (also known as c.1894G>A), located in coding exon 6 of the CASR gene, results from a G to A substitution at nucleotide position 1894. The glycine at codon 632 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
