Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000222.3(KIT):c.859G>T (p.Val287Leu), citing Ambry Variant Classification Scheme 2023: The p.V287L variant (also known as c.859G>T), located in coding exon 5 of the KIT gene, results from a G to T substitution at nucleotide position 859. The valine at codon 287 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
