This variant lies in the ATP7B gene (transcript NM_000053.4) at coding-DNA position 3235, where T is replaced by C; at the protein level this means replaces cysteine at residue 1079 with arginine — a missense variant. Submitter rationale: This variant has been observed in individual(s) with clinical features of Wilson's disease (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant disrupts the p.Cys1079 amino acid residue in ATP7B. Other variant(s) that disrupt this residue have been observed in individuals with ATP7B-related conditions (PMID: 21796144), which suggests that this may be a clinically significant amino acid residue. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (ExAC no frequency). This sequence change replaces cysteine with arginine at codon 1079 of the ATP7B protein (p.Cys1079Arg). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and arginine.
