This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 8927, where G is replaced by A; at the protein level this means replaces serine at residue 2976 with asparagine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function.
