NM_000393.5(COL5A2):c.905G>T (p.Arg302Leu) was classified as Uncertain significance for Ehlers-Danlos syndrome, classic type, 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with leucine at codon 302 of the COL5A2 protein (p.Arg302Leu). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and leucine. This variant is present in population databases (rs545574407, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with COL5A2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
