This variant lies in the IFT140 gene (transcript NM_014714.4) at coding-DNA position 1837, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 613 with asparagine — a missense variant. Submitter rationale: Variant summary: IFT140 c.1837G>A (p.Asp613Asn) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251486 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1837G>A has been reported in the literature in one individual affected with severe oligozoospermia, asthenozoospermia, and teratozoospermia (OAT) without other physical abnormality (Wang_2019). The report does not provide unequivocal conclusions about association of the variant with Retinitis Pigmentosa. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
