This variant lies in the STAT2 gene (transcript NM_005419.4) at coding-DNA position 2236, where G is replaced by C; at the protein level this means replaces aspartic acid at residue 746 with histidine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid with histidine at codon 746 of the STAT2 protein (p.Asp746His). The aspartic acid residue is moderately conserved and there is a moderate physicochemical difference between aspartic acid and histidine. This variant is present in population databases (rs760470820, ExAC 0.002%). This variant has not been reported in the literature in individuals with STAT2-related conditions.
