NM_000057.4(BLM):c.2489C>A (p.Thr830Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T830K variant (also known as c.2489C>A), located in coding exon 11 of the BLM gene, results from a C to A substitution at nucleotide position 2489. The threonine at codon 830 is replaced by lysine, an amino acid with similar properties. This alteration was identified in male diagnosed with breast cancer (Rizzolo P et al. Int J Cancer, 2019 07;145:390-400). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
