NM_002408.4(MGAT2):c.206A>G (p.Asn69Ser) was classified as Uncertain significance for MGAT2-congenital disorder of glycosylation by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with serine at codon 69 of the MGAT2 protein (p.Asn69Ser). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and serine. This variant is present in population databases (rs376612891, ExAC 0.02%). This variant has not been reported in the literature in individuals with MGAT2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0").
