NM_203447.4(DOCK8):c.250G>A (p.Gly84Arg) was classified as Uncertain significance by Genetic Services Laboratory, University of Chicago: DNA sequence analysis of the DOCK8 gene demonstrated a sequence change, c.250G>A, in exon 3 that results in an amino acid change, p.Gly84Arg. This sequence change does not appear to have been previously described in individuals with DOCK8-related disorders. This sequence change has been described in the gnomAD database with a frequency of 0.0014% (dbSNP rs1318020471). The p.Gly84Arg change affects a moderately conserved amino acid residue located in a domain of the DOCK8 protein that is known to be functional. In-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL) provide contradictory results for the p.Gly84Arg substitution. Due to insufficient evidences and the lack of functional studies, the clinical significance of the p.Gly84Arg change remains unknown at this time.
