Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000075.4(CDK4):c.644G>C (p.Cys215Ser), citing Ambry Variant Classification Scheme 2023: The p.C215S variant (also known as c.644G>C), located in coding exon 5 of the CDK4 gene, results from a G to C substitution at nucleotide position 644. The cysteine at codon 215 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
