Uncertain significance for Intellectual disability, X-linked 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001111125.3(IQSEC2):c.3935C>T (p.Ser1312Leu), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with IQSEC2-related conditions. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This sequence change replaces serine with leucine at codon 1312 of the IQSEC2 protein (p.Ser1312Leu). The serine residue is weakly conserved and there is a large physicochemical difference between serine and leucine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
