Uncertain significance for Developmental and epileptic encephalopathy, 26 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004975.4(KCNB1):c.752C>T (p.Ser251Leu), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with KCNB1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with leucine at codon 251 of the KCNB1 protein (p.Ser251Leu). The serine residue is highly conserved and there is a large physicochemical difference between serine and leucine. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
