Uncertain significance for ALG2-congenital disorder of glycosylation; Congenital myasthenic syndrome 14 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_033087.4(ALG2):c.30del (p.Ser11fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ser11Argfs*37) in the ALG2 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in ALG2 cause disease. This variant is present in population databases (rs763575022, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with ALG2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1016801). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
