This variant lies in the MMUT gene (transcript NM_000255.4) at coding-DNA position 227, where G is replaced by A; at the protein level this means replaces arginine at residue 76 with lysine — a missense variant. Submitter rationale: This sequence change replaces arginine with lysine at codon 76 of the MUT protein (p.Arg76Lys). The arginine residue is moderately conserved and there is a small physicochemical difference between arginine and lysine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with features of methylmalonic acidemia (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant.
