This variant lies in the LAMB2 gene (transcript NM_002292.4) at coding-DNA position 4919, where A is replaced by G; at the protein level this means replaces tyrosine at residue 1640 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tyrosine with cysteine at codon 1640 of the LAMB2 protein (p.Tyr1640Cys). The tyrosine residue is weakly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with LAMB2-related conditions.
