This variant lies in the PTCH1 gene (transcript NM_000264.5) at coding-DNA position 1728, where G is replaced by T; at the protein level this means replaces glutamine at residue 576 with histidine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This sequence change replaces glutamine with histidine at codon 576 of the PTCH1 protein (p.Gln576His). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and histidine.
