Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003001.5(SDHC):c.251T>G (p.Leu84Arg), citing Ambry Variant Classification Scheme 2023: The p.L84R variant (also known as c.251T>G), located in coding exon 5 of the SDHC gene, results from a T to G substitution at nucleotide position 251. The leucine at codon 84 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
