Uncertain significance for Nephronophthisis 18 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016122.3(CEP83):c.1273C>G (p.Gln425Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with glutamic acid at codon 425 of the CEP83 protein (p.Gln425Glu). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and glutamic acid. This variant is present in population databases (rs370718016, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with CEP83-related conditions. ClinVar contains an entry for this variant (Variation ID: 1015337). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
