NM_001363.5(DKC1):c.41A>G (p.Lys14Arg) was classified as Uncertain significance for Dyskeratosis congenita by Sema4, Sema4, citing Sema4 Curation Guidelines. This variant lies in the DKC1 gene (transcript NM_001363.5) at coding-DNA position 41, where A is replaced by G; at the protein level this means replaces lysine at residue 14 with arginine — a missense variant. Submitter rationale: The DKC1 c.41A>G (p.K14R) variant has not been reported in the literature to our knowledge. It was observed in 8/19148 chromosomes of the African/African American (AFR) subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID 1014953). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
