This variant lies in the RELN gene (transcript NM_005045.4) at coding-DNA position 1464, where T is replaced by G; at the protein level this means replaces asparagine at residue 488 with lysine — a missense variant. Submitter rationale: This sequence change replaces asparagine with lysine at codon 488 of the RELN protein (p.Asn488Lys). The asparagine residue is weakly conserved and there is a moderate physicochemical difference between asparagine and lysine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with RELN-related conditions.
