Uncertain significance for Orofacial cleft 6, susceptibility to; Van der Woude syndrome; Popliteal pterygium syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006147.4(IRF6):c.1270T>G (p.Ser424Ala), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with IRF6-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces serine with alanine at codon 424 of the IRF6 protein (p.Ser424Ala). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and alanine.
