Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001903.5(CTNNA1):c.265G>A (p.Glu89Lys), citing Ambry Variant Classification Scheme 2023: The p.E89K variant (also known as c.265G>A), located in coding exon 2 of the CTNNA1 gene, results from a G to A substitution at nucleotide position 265. The glutamic acid at codon 89 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
