Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.5354C>A (p.Thr1785Lys), citing Ambry Variant Classification Scheme 2023: The p.T1785K variant (also known as c.5354C>A), located in coding exon 39 of the POLE gene, results from a C to A substitution at nucleotide position 5354. The threonine at codon 1785 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
