This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 5095, where T is replaced by A; at the protein level this means replaces tyrosine at residue 1699 with asparagine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change replaces tyrosine with asparagine at codon 1699 of the FBN1 protein (p.Tyr1699Asn). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and asparagine. This variant has been observed in individual(s) with acromicric dysplasia (Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 1014036). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant disrupts the p.Tyr1699 amino acid residue in FBN1. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 21683322). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
