NM_024426.6(WT1):c.676A>C (p.Thr226Pro) was classified as Uncertain significance for Wilms tumor 1; Drash syndrome; 11p partial monosomy syndrome; Frasier syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces threonine with proline at codon 221 of the WT1 protein (p.Thr221Pro). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and proline. This variant has not been reported in the literature in individuals with WT1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
