This variant lies in the APOB gene (transcript NM_000384.3) at coding-DNA position 9600, where C is replaced by G; at the protein level this means replaces serine at residue 3200 with arginine — a missense variant. Submitter rationale: This sequence change replaces serine with arginine at codon 3200 of the APOB protein (p.Ser3200Arg). The serine residue is weakly conserved and there is a moderate physicochemical difference between serine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with clinical features of familial hypercholesterolemia (Invitae).
