Uncertain significance for X-linked distal spinal muscular atrophy type 3; Cutis laxa, X-linked; Menkes kinky-hair syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000052.7(ATP7A):c.1876G>C (p.Gly626Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with arginine at codon 626 of the ATP7A protein (p.Gly626Arg). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and arginine. This variant is present in population databases (rs782396059, ExAC 0.004%). This variant has not been reported in the literature in individuals affected with ATP7A-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ATP7A protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
