ALK functions as a receptor tyrosine kinase and fusion events typically retain the kinase domain of the protein, such as in this tumor, resulting in constitutive activation of ALK. PRRC2B is a proline-rich coiled-coil protein with unknown function, though single case reports of fusions involving the 5-prime portion of PRRC2B have been described. The PRRC2B-ALK fusion is expected to result in constitutive activation of ALK and is consistent with immunohistochemical evidence of ALK positivity (cytoplasmic) in the tumor.
