Uncertain significance for Ornithine aminotransferase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000274.4(OAT):c.650G>A (p.Arg217His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with histidine at codon 217 of the OAT protein (p.Arg217His). The arginine residue is moderately conserved and there is a small physicochemical difference between arginine and histidine. This variant is present in population databases (rs774853325, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with OAT-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
