This variant lies in the NBN gene (transcript NM_002485.5) at coding-DNA position 2215 through coding-DNA position 2216, deleting 2 bases; at the protein level this means shifts the reading frame starting at leucine residue 739, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Leu739Cysfs*2) in the NBN gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 16 amino acid(s) of the NBN protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NBN-related conditions. Algorithms developed to predict the effect of variants on protein structure and function are not available or were not evaluated for this variant. Experimental studies have shown that this premature translational stop signal affects NBN function (PMID: 15758953).
