Uncertain significance for Melanoma, cutaneous malignant, susceptibility to, 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002386.4(MC1R):c.668G>A (p.Arg223Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with glutamine at codon 223 of the MC1R protein (p.Arg223Gln). The arginine residue is moderately conserved and there is a small physicochemical difference between arginine and glutamine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has been observed in individual(s) with melanoma and basal cell carcinoma (PMID: 16645598, 15998953, 18067130). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
