Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.3311G>A (p.Ser1104Asn), citing Ambry Variant Classification Scheme 2023: The p.S1104N variant (also known as c.3311G>A), located in coding exon 20 of the DICER1 gene, results from a G to A substitution at nucleotide position 3311. The serine at codon 1104 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
