This variant lies in the EFHC1 gene (transcript NM_018100.4) at coding-DNA position 1886, where T is replaced by G; at the protein level this means replaces isoleucine at residue 629 with serine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with serine at codon 629 of the EFHC1 protein (p.Ile629Ser). The isoleucine residue is moderately conserved and there is a large physicochemical difference between isoleucine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with EFHC1-related conditions.
