Uncertain significance for Nephronophthisis 15 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014956.5(CEP164):c.829G>T (p.Gly277Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with cysteine at codon 277 of the CEP164 protein (p.Gly277Cys). The glycine residue is weakly conserved and there is a large physicochemical difference between glycine and cysteine. This variant is present in population databases (rs772164945, ExAC 0.005%). This variant has not been reported in the literature in individuals affected with CEP164-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The cysteine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
