This variant lies in the SQSTM1 gene (transcript NM_003900.5) at coding-DNA position 1060, deleting one base; at the protein level this means shifts the reading frame starting at glutamine residue 354, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Gln354Serfs*24) in the SQSTM1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 87 amino acid(s) of the SQSTM1 protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with SQSTM1-related conditions.
