Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.397G>T (p.Asp133Tyr), citing Ambry Variant Classification Scheme 2023: The p.D133Y variant (also known as c.397G>T), located in coding exon 2 of the BLM gene, results from a G to T substitution at nucleotide position 397. The aspartic acid at codon 133 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
