Uncertain significance for Dilated cardiomyopathy 1CC; Hypertrophic cardiomyopathy 20 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_144573.4(NEXN):c.794A>C (p.Gln265Pro), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with NEXN-related conditions. This sequence change replaces glutamine with proline at codon 265 of the NEXN protein (p.Gln265Pro). The glutamine residue is highly conserved and there is a moderate physicochemical difference between glutamine and proline. This variant is not present in population databases (ExAC no frequency).
