This variant lies in the CSRP3 gene (transcript NM_003476.5) at coding-DNA position 49 through coding-DNA position 55, deleting 7 bases; at the protein level this means shifts the reading frame starting at valine residue 17, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change is expected to alter the c-terminus of the CSRP3 protein (p.Val17Metfs*189). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 178 amino acid(s) of the CSRP3 protein and extend the protein by 10 additional amino acid residues. This variant is present in population databases (rs776444956, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with CSRP3-related conditions. ClinVar contains an entry for this variant (Variation ID: 1009234). This variant disrupts a region of the CSRP3 protein in which other variant(s) (p.Cys150Tyr) have been determined to be pathogenic (PMID: 23396983, 25351510, 33035702). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
