NM_001148.6(ANK2):c.9404G>T (p.Gly3135Val) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G3135V variant (also known as c.9404G>T), located in coding exon 38 of the ANK2 gene, results from a G to T substitution at nucleotide position 9404. The glycine at codon 3135 is replaced by valine, an amino acid with dissimilar properties. This exon is expressed solely in brain (Mohler PJ et al. Circulation. 2007;115(4):432-41). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
