Uncertain significance for Deficiency of aromatic-L-amino-acid decarboxylase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001082971.2(DDC):c.1392G>C (p.Trp464Cys), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with DDC-related conditions. This variant is present in population databases (rs372970018, ExAC 0.003%). This sequence change replaces tryptophan with cysteine at codon 464 of the DDC protein (p.Trp464Cys). The tryptophan residue is highly conserved and there is a large physicochemical difference between tryptophan and cysteine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
