This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 105863, where T is replaced by G; at the protein level this means replaces isoleucine at residue 35288 with serine — a missense variant. Submitter rationale: This variant is located in the M band of TTN (PMID: 25589632). Variants in this region may be relevant for neuromuscular disorders, but have not been definitively shown to cause cardiomyopathy (PMID: 23975875). This sequence change replaces isoleucine with serine at codon 35288 of the TTN protein (p.Ile35288Ser). There is a large physicochemical difference between isoleucine and serine.
